ANALYSIS OF THE GARLIC LANDRACES IN THE WEST OF ROMANIA USING THE FLOWCYTOMETRY METHOD

ABSTRACT

For to determinate the DNA content and analysis of cell distribution in different phases of the cell cycle, flow cytometry was used. The exact measurements of a large number of nuclei by flow cytometry, fluorescently labeled by passing a laser beam (excitation), which marks each individual nucleus by fluorescence, allows a rapid estimation of the relative nuclear content of the DNA of the analyzed plants . Approximately 0.5 cm2 of foliar tissue is harvested, crushed very well, followed by incubation of the sample at room temperature after which the mixture is poured into a filter by adding a coloring solution, the samples are incubated in the dark for at least 1 hour before to analyze at the flow cytometer. The samples thus prepared are stable for 12 hours at 4 ° C. For this study six garlic landraces from two counties in Romania were used, the number of cells analyzed at Flowcytometer varied between 5000 and 10000, which is sufficient to analyze the distribution of cells in the divisional phases. No abnormal evolution was noted in the six garlic landraces analyzed, which revealed the genome’s stability, the ability to adapt to environmental conditions.
