USE OF IDEXX QUANTI-TRAY SYSTEM FOR RAPID ASSESSMENT OF FAECAL INDICATOR BACTERIA (FIB) IN COASTAL BLACK SEA SURFACE WATERS

ABSTRACT
Sanitary quality survey of seawater using faecal indicator bacteria (FIB), specifically Total Coliforms (TC), Faecal Coliforms (FC), Escherichia coli and Faecal Streptococci (FS) is a common practice in populated coastal areas around the world. This study provides the first investigation of FIB in coastal Black Sea waters by means of the IDEXX Quanti-Tray 2000 most probable number system (Colilert-18 and Enterolert-E).The study was conducted in spring 2013 in six sampling points along the Romanian coastline, covering both unpolluted and polluted areas. The obtained results were compared with data resulted through the traditional multiple-tube fermentation (MTF) method for measuring indicator bacteria levels, in order to evaluate the suitability of IDEXX Quanti-Tray system in assessment of FIB in Black Sea waters. We found that Colilert-18 and Enterolert-E yielded in less than 24 hours comparable results to MTF method in detecting indicator microorganisms in the Black Sea waters. The IDEXX results were highly correlated with MTF results for all three indicators regardless of whether the samples were taken along estuarine, open shoreline or near a sewage runoff (R2= 0.99 for TC, R2= 0.98 for E.coli/FC, and R2 = 0.80 for FS). Our study indicate that IDEXX methods offer several advantages over the traditional methods for detecting faecal contamination in the Black Sea waters and could be recommended as standard methods for the marine water quality monitoring program.
